Has anyone done this route, and how long did it take?

I've not done it all in one go, but have driven Barcelona-Perpignan and Perpignan-Calais many times (my parents live in Perpignan). If (in France) you travel via Beziers, Viaduc de Millau, Clermont, Orleans, and Paris, you'll need about 13-14hrs actual driving time Barcelona-Calais.

In case it helps, I'll offer some data from the ViaMichelin route planner:

The journey distance is given as 823 miles, with nearly all of that (806 miles) being on motorways. The total driving time is given as 12 hours 11 minutes (including 11 hours 35 minutes of motorway driving).

The total cost is estimated at £138.31, with £81.76 of that being petrol and £56.55 being toll charges.

Looking at those figures you could probably do it in a single, long, tiring day with two drivers. It would be more comfortable across two days.

A single driver could also manage it over two, very tiring days but it would probably be better to spread the journey over three days.

Chris... I've actually got to drive Valencia to Calais with 1 overnight stop. I tried googling Valencia to Calais, but there wasn't much coming up, and what did, conflicted greatly. My plans are changing by the day, but I'm thinking of leaving at about 5 or 6am on Saturday next week, and I'm hoping to catch the Eurotunnel in Calais at around 2-4pm on Sunday, stopping overnight in either Bourges or Orleans, depending on how far I get.
assuming it's 5hr from valencia to the border, it'll take you about 13hr to reach orleans (or around 12 to get to bourges).

on the other hand it's only just over 4hr from orleans to calais, so it might be less knackering to stop over further south, say clermont or montlucon.

Is that allowing for coffee breaks?

I also have a dog so need to get to Calais 1-2 hours before departure to allow for his passport, etc. to be checked.
the times shown assume an average of 60mph/100km-h, which I've found allows for fuel & loo stops but not much else.
