mileagewise it seems Manchester is far closer to Birmingham

Try Warwick - 117 miles from Manchester and 113 miles from Southampton according to the AA.
The Cotswolds. Plenty of nice places to visit there.
If you go into Google maps it is easy to work it out yourself.

Find Manchester and RIGHT click then select "Directions from here" then find say "Warwick" and again right click and select "Directions to here".

That gives you the distance in miles and the driving time.

Do the same from Southampton and play around with the various "routes" to find a central one.
Milton Keynes. Now there's a place
Paris, about the same flight time from both, but much more fun!
Are you both driving, or going by train or coach? That might make a difference to where we suggest, how many changes there might be on public transport.
According to Google maps it is 234 miles from centre of Manchester to centre of Southampton. 4 hours driving time.

This is down the M6, round Birmingham, then down M40 then A34 to Southampton.

So you want something about 2 hours driving time from each place.

As has been said, Warwick fits the bill (it is on the M40 as well so easy to get to).

If you want something larger nearby then maybe Stratford upon Avon (lovely town with lots to do), or Coventry (not such a lovely town).
Met on the internet and wanna find somewhere halfway to meet?
We tend to have get togethers in Stratford for the Lancashire, Yorkshire, Wales and South Coast friends.
The midpoint is on the International Dateline to the south-east of New Zealand if you choose to go the long way around.
Southhampton to Man approx 41/2 hrs, half way, Warwick.
Cork is roughly midway between the two and is a lovely place to visit at this time of year
