As far as I can make out, you can't fly direct from anywhere in the UK - just from Chennai and Kolkata in India.
There's no international airport within the Andaman Islands. All air travel has to be via Chennai or Calcutta. See here:
http://www.andamanisland.com/reach.htm

There are also no direct flights from Manchester to any Indian mainland destination. See here:
http://www.manchesterairport.co.uk/app/AllDest inations

Thus, you'd have to commence your journey from Manchester by flying to a suitable international hub, such as Heathrow or Amsterdam.

BA don't offer First Class between Manchester & Chennai. The best they can do is Economy for the domestic leg to Heathrow, followed by Club Class for the international flight. For a random date I've just checked, they quote a total of 6396.60 for 2 people.

Indian Airlines quote US$488.40 for two people, travelling Executive Class, between Chennai and Port Blair.

Foreign (non-Indian) nationals need an Indian government visa and, (available on arrival), an entry permit. The permits are valid for 30 days. I would expect that you would also need to show an onward or return ticket on arrival.
First Class fares from Manchester appear to be unavailable, presumably because some sectors of the route will not have First Class seating in the planes.
For fares, enter details here and you'll get a list of alternative airlines and prices.
(Kolkata, of course, used to be called Calcutta).
