I have all clues solved. I have the quotation inserted. Please give me some clue as to what might go in the 8 isolated squares and as to what the 3 letter word might be.

Look at the diagonals!

I'm sort of stuck on the three letter word, but I have an idea what it might be based on a couple of anomalies in the clues and the "two knights moves" bit. Can anyone tell me if it is a word meaning understand/work out? Seems to fit, but I'm not sure it sums up the theme...
I don't get the 3 letter word. Why the 2 extra s's?

The grid is missing 2 symmetrical bars, 1 after the 3-letter 11 across entry & the other on right side of square containing 35. The number 35 must be moved 1 square to the right.

Alilloyd, if you mean SEE, then no it's not. There is a particular reason why MynoT has orientated the isolated squares with NW pointing directly up, and it relates to a specific (different) quotation regarding the main theme of the play in question. The 3-letter word is an adjective derived from this theme.
MLN ~ the missing bars you mention, and the 35 to be moved ~ are you saying these are mistakes by the setter?

A relative in another city has phoned me for help with the 3-letter word but I don't have the crossword. Could it be described as play or behave?

Your diagonals are correct Ripper, but you need to ignore the central cell, ie start in the 4 inner isolated squares to 'find directions out'.

For the purposes of this puzzle a knight's move is 2 up and 1 to the right - you just need to work out where to start, and my advice is don't wander too far from the centre (see my first post above)

Like you guys I finished everything but the 3 letter word quite quickly. I think I've found this missing "insane" word but am a bit thrown by its direction as as the quotation that led me to it implies it should go NNW - ie I thought I'd find it with the first 2 letters going up and the 3rd diagonally to the left of them. However- the manifestation I have found has 2 letters north of the 3rd, which is on row below, to the west of them. Have I missed something or is this it?
As I mentioned above, for the purposes of this puzzle a knight's move is 2 up and 1 to the right. Had the 3-letter word run directly up, this would have been NW - it therefore runs north of NW = NNW (approximately)
