I recently purchased an Omega Super lighter MFGCO -

Thus far been I've been unable to find a definitive answer as to how to refill it. On the underside are two screws - The first has a spring attatched (interior) that holds a tiny piece of flint and the second screw opens to a casing of, I think cotton wool.

Do I need to pour lighter fluid onto the wool, buy a new flint or both...or neither?

The inner part,with the flint and rotary striker should pull out,you then pour lighter fluid onto the cotton at the base,refit and, viola,flames.
If the inner doesn't pull out, pour the lighter fuel into the hole that shows the cotton wool.
If it's like a zippo lighter then it's as paddiez and chakka has said, here's a video of how.

No problem at all.
this lighter works just like a zippo, you pour the lighter fluid in the larger screw hole, the other will hold a flint you can actually use zippo flint in it as well, (i have tested this), but if you can click it and a spark comes then you don't need new flint. fill the cotton with lighter fluid until fully saturated, don't over fill you would just be wasting fluid.
