Why is it difficult to analyse glucose directly using HPLC with UV detection?

Well, assuming that you need another short answer, it's because glucose absorbs virtually no UV light.

Yes, that's right. This problem occurs in the majority of saccharides.

This is also why other methods are used for other molecules lacking chromophores such as amines and aldehydes.
