I need to get from New Yrk JFK Airport to a Hotel near Penn Station - can anybody tell me if it is quicker to use to subway or is there a bus?

There is no easy way on the subway to et to the city from JFK. It really is not a good idea to do it this way. Not sure about buses either, but I'm sure they do run this route.

I would recommend getting a cab from the airport, there is always a 'guard' on the cab line and they organise the taxi queue and make sure you get into a cab safely etc. There is a set fare for the trip from the airport to Manhattan, I think it is around $45, well worth it and you get a good view of the skyline on your way in, far less hassle then having to sort out tickets for buses etc and not being sure where to get off.
You can do it by train from JFK Terminal 4 Station to Penn Station, but you have to change services at Jamaica Station.
I used this bus service 2 years ago and they were great. Just walked out the terminal at JFK and there was a guy standing outside shouting if anyone wanted tickets (it seemed very unofficial) but it was great and cheap. Buses were bang on time there and back.

But it takes up to 2 hours!!!!!
Well i was close, its the though that counts.

I got this from JFK to Port Authority (don't remember it taking that long though) but you get a great view of the city, cross some of the bridges and also get a mini tour of Manhatten as it drives through, so i thought it was well worth it.
Catch a cab for fixed rate $45 plus tolls and tips.
We went about 18 months ago and booked a shuttle bus in advance for travelling from airport to Pennsylvania hotel - it was much cheaper than a taxi and was very straightforward when we arrived.
