Try not to upset your staff and make sure that 2 people know all the passwords. Its common practice in I.T. when people resign to show them the door straight away. Not a sure fire was to stop this but helps. Theres simply no point having a password system which can be broken so you a bit stuck, can you export the data and the rebuild the database. You could threaten to take the former employee to court for crimial damage and see if he cracks.
Try holding down the shift key when you enter the database. Failing that there are programmes out there that can find lost passwords. Here is one for example

They are all pretty pricey though - as one would expect.
The one rinkytink suggests is avalaible to try for free well worth a go for free.
Does it ask you for a username/password to access the database? If yes: use the administrators account and see if you can adjust it. An alternative is to retore the database from a backup created before the permissions were changed. If not then the permission may have been set at the NTFS/share level. This is very easy to reset if you know the administrators account password.
