Do you mean best mode of travel or driving directions? If driving it is well signposted all the way - little bit of A30 and M5 round Exeter then A38 all the way until Plymouth exit where you follow the signs to city centre. Takes about an hour.
Taxi to Exeter StDavids (takes about 15 mins). Train to Plymouth takes an hour.

Thank you Prudie an Count-A -Long.
If time is not a factor, but cost is, there is a bus ( No.56, or No. 379 on Sunday) from the airport terminal to Exeter St David's Station every hour, takes about 30 minutes during the week, 25 minutes on Sunday. Likely to be cheaper than taxi.
