... in 342 AD, the Emperor Constantine passed a law forbidding ..:

Same sex marriage
Oral sex
Anal sex

I'm not saying that I'd actually want it in the butt. That's just a personal preference thing. But, you know ...

Was this the earliest example of legislation that discriminated on the grounds of sexual orientation?

The Bible must have had it before 342AD , it is even in the Old Testament, that was 100s or even 1,000s of years before Christ. Koran was not written until 600 years or so after the New Testament. Didn't know blow jobs were illegal anywhere though.

those are bans on actions, not orientation. They'd have applied to straight people too. Not that straights would be likely to marry but they have been known to indulge in the other practices, or so I am told.
