According to this site, it was Playschool on 21st April 1964:

Yes, Play School, only because the opening night was affected by a power cut at TV Centre.

They did manage to get a news bulletin out on the opening night, so if you want to be picky, technically the News was the first programme on BBC2.

21 April 1964 First programme broadcast on BBC2 - Play School at 11.00am.
