Audio sounds have pitch, which can be defined as a frequency eg A above middle is 440Hz, usual concert pitch.
So that pitch can easily defined as a midi note along with its note on/off information
theres no secret. its acoustic mathematics, physics of sound and programming skills

monophonic sounds are much easier to convert to midi, polyphonic not so.
Companies like Celemony have made big inroads in polyphonic audio to midi software Melodyne with DNA, i use it all the time.
