Phrase you have there may mean something to do with jabbing a rooster in the derriere, (rather than its sexual organs) - ie, something which wakes a rooster up in the early morning; probably a slang term.

I'm not familiar with the film, but I should imagine the phrase is a reference to the tendency for men to have early-morning erections. Would that fit the movie-script?

I should have added above, that such phenomena are more often the result of the full bladder affecting blood-flow to the penis rather than anything genuinely erotic!

It's not a sexual reference, but 'prick' in the sense of obnoxious, like a rooster fighting for attention. In fact, Celene explains it herself when Jesse questions her:

Jesse: I was a rooster prick?
