Does Vatican City count as a country in its own right, or is it just a region of Italy?

Time-Out treasure hunt, clue number29 concerning the Globe Theatre
The iron gates were made from individual characters cast by 130 blacksmiths in 1997. Only the Queen has gone through the gates. One of the characters only is burnished and multi-coloured, and question is 'What is the source of magic of this character ? I have viewed the gate, and it looked like 'nails' to me, but wondered whether was magic- mushrooms? would really appreciate help, please
