I've solved most of this puzzle apart from the extreme bottom right hand corner. I also can't find enough misprints so am hoping that's where they're hiding! Any tips greatly appreciated!!

Which ones are you actually missing? I think I've done the bottom left, but am missing 1a & 2d ..... and although I know what I'm looking for I've also failed to locate some misprints.

I'm stuck on top left and the theme
I've got the two start letters
Stuck on RH top corner too. 2d I have ??ll. 3d I think is "trait". Gut feeling for 1a is "ENT". but cannot qualify any of these speculated answers with the clues given. Completely stuck on 43d, have ca?. Any hints on the starting points would be helpful.

43down is CAT (sick according to chambers) T is 160 (again Chambers)

Thanks for 43 d. This one is a right burger. Did wonder if 2d was cull, "will" being altered to "kill" cannot qualify that though. 29a, is it gorgon? That would make 12 d, which I've got most of the letters for, a rather odd word. Maybe it's a computing term?
Absolutely no idea what the theme is.
We need K to come to the rescue.
Can't see it beong cull, 'cos we need a Note before.
29ac is right
29d 'does what it says on the tin' look under GO in the dictionary (it took me ages - so I'm making you work on that one!!!!)
If ENT is right then 2 down should be N-LL

Thanks Kettledrum. The ones I'm not sure of are 39A, 40D and 43D. I thought 39 might be COCA and 43 might be CEP but I've no idea why so I've probably misled myself (again!). For 2D I'm torn between TELL (as in Willian Tell) & FELL.

Thanks Kettledrum! I like those answers much better than mine! For Judypark - I agree with your 29A. In my Chambers the answer to 29D is hyphenated after the first 2 letters, and means Pipe cleaner. I also like NOLL for 2D and am thinking perhaps it's an old spelling for knoll, meaning Hill.
I think 2d is fall (wilt).

The theme (again hypothesis) is a bingo number (house call) which may indicate shape of lines!

Like everybody else though, still stuck on SE corner.

definitely not = no
hollow "the" = te
note is a synonym for stem
Thanks ulysses, now completed the grid but am not sure of the starting points of the two lines. Also not sure of what I'm looking for, is it something along the lines of "two little ducks" or do I need to look for something more subtle?
I also have a completed grid (but unsure about a couple of them ) and most of the misprints which do in fact represent a house call, but a bit higher up than the one you have suggested. I think that I also have one of the starting letters - the "C" in 31 ac but, as yet, I cannot work out the required shapes.
Hi Ulysses100, I am in exactly the same position as you. I suspect that the lines may be in the shape of the two figures suggested by the call described by the misprints but can’t see that the C in 31a is an obvious starting point for anything logical.
Hi I am in same position complete grid, two ??? ?????? as the house call, and O from 23a and C from 31a as starting points but now staring at grid and getting nowhere.
Hoist the flags and let joy be unbridled, finally cracked it. Mad max's break down is correct so that should narrow it down a bit. Without spoiling it for anybody, but for those who now fear for their sanity. Top left hand corner. from the B in 17a, read the letters in an anti-clockwise direction, similarily follow the O of 23a in a clockwise direction, the right hand side can be identically solved in a symmetrical manner.
