I think that he intended it to be humorous, I don't imagine he intended any insult, but it's crass and it's childish and it's not funny. I think it's unfortunate but you do have to balance intention vs result with remarks like that and if the lady in question felt 'humiliated, shocked and enraged', then I'm simply happy for her that that's the biggest problem she's ever encountered.
Casual racism is an issue but particularly with some older people it's unintentional and we need to man up and not be quite so touchy when no offence is intended. It's fine to point it out, it's fine to correct it, but don't give your power away and be 'humiliated, shocked and enraged' by some muppet who despite the expensive education doesn't understand how to behave properly.

I shook the prince’s hand with my right hand. In my other, I was holding a copy of an anthology, We Mark Your Memory: Writing from the Descendants of Indenture, in which I have an essay published. I told him that my mother was born in Guyana and that the anthology had collected hidden histories of indenture.
“And where are you from?” asked the prince.
“Manchester, UK,” I said.
“Well, you don’t look like it!” he said, and laughed. He was then ushered on to the next person.

It's the "don't look like it" bit, isn't it? She's not white, so she doesn't look British.
