We are both college freshman and I talked to him twice. A couple of days ago we said hello, how are you. And I asked him to hang out but unfortunately he had a night class and said we can probably hang out this weekend. But I never really see him and he’s seems shy. Should I knock on his door & try again? Or leave a note for his door ?

Since you are in America
And this is a UK site , I'm not sure what to advise, maybe going over to him in the canteen where you'd eat may be better than knocking on his door, perhaps taking your tray and asking if you could join him?
why would living in America affect any advice given ?
Because I'm not familiar how old a 'freshman' is ?
And that would play a part in any advice I could give
I'd leave a note, something simple like - Fancy a coffee? and your contact info. It's up to him then if he takes you up on your offer, and you can either take it from there, or not.
Or tell him you fancy him something rotten

If he reciprocates , you're on
If he smiles and says he's too busy with his studies right now and don't need any distractions - then move on

À freshman is a first year student...hence 'fresh'. So roughly 18 or so.

Wait to bump into them again and set up a coffee date or something. A note could look stalker-y, or fall into the wrong hands and cause embarrassment.
If he is shy then I am not sure that being upfront eg asking him out, making a date etc would be the best thing...you may well scare the living daylights out of him. I am assuming that you know little about his personal life ...he may have a girlfriend at home, he may be gay, he may be dealing with some difficult issues in his family...Sooooo my advice would be to get to know him as a friend first, bump into him, go for a coffee (on a really casual basis...you know 'I am just off for a coffee ..want to come') and see how you relationship developes...or not , as the case may be.
