Just how brave are you feeling?
Pakistan shares borders with India, China, Afghanistan and India. Entry via India is illogical because you'd have to go via China to get there. Entry from Afghanistan is near suicidal because of the areas which are effectively beyond the control of the respective governments (where terrorists, war lords and various rebel groups control the area and have a nasty habit of kidnapping travellers). Iran offers a possible route but, once again, there's a high risk of nasty consequences if you venture into the border areas. Entry from China might be possible but that would mean travelling through the Hindu Kush, which is inhospitable for many reasons. (There are rebel-held areas and driving through the high mountain passes should probably only be attempted in a convoy of 4-wheel drive vehicles, preferably with an armed escort).
The straight-line distance between London and Islamabad is about 3800 miles. By road it's much further. Most routes would probably take you via Ukraine. To plan a route for the first part of the journey, enter 'London' and 'Kiev' here:

It's about 1500 miles. A driver who's used to all-day driving (e.g. a trucker) might be able to do it in 3 days but most drivers would probably need 4 days to stay alert while driving.
Once you've got to Kiev, you've only covered about a third of the driving distance and things get much slower from there on. I reckon it would probably take about 3 weeks to get to Pakistan. Also, as stated, I can't see any safe way of getting there without a specialised vehicle for the mountainous areas, a suitable back-up plan (i.e. a support vehicle and satellite radio links) and an armed escort for the 'troubled' areas.
